Understanding USP Column Equivalence (USP <621>)

When a specific HPLC column used in a validated method is unavailable or discontinued, USP General Chapter <621> Chromatography allows the use of an "equivalent" column. The primary requirement is that the alternative column must share the same L-Classification (L-Code) and be capable of meeting System Suitability requirements.

Step 1

Identify Your Column: Select the manufacturer and specific column name you are currently using from the dropdowns above.

Step 2

Verify Dimensions: Ensure the Length, Diameter, and Particle size match your current method. While USP <621> allows some adjustments to dimensions for isocratic methods, sticking to the original specs minimizes re-validation work.

Step 3

Compare Alternatives: The tool lists equivalent columns from both premium global brands (like Waters, Agilent) and high-quality value manufacturers (like Welch, GL Sciences, Kromasil). This allows you to balance performance needs with budget constraints.

Common USP Codes

  • L1 (C18): Octadecyl silane bonded to porous silica or ceramic microparticles.
  • L7 (C8): Octyl silane bonded to totally porous silica particles.
  • L11 (Phenyl): Phenyl groups bonded to porous silica particles.
  • L3 (Silica): Porous silica particles (Normal Phase).
